Hi there, James! We recommend closing the bag with a clip of some kind so that you don't lose any frosting or filling out of the back, especially if you need to put the bag down. You're certainly welcome to close the bag with a simple twist if you'd prefer though! Just be sure not to twist the end of the bag too much for the pressure will make the frosting or filling come out of the tip. A pastry bag can absolutely be filled while you're holding it with the opening folded over your hand, but if you're new to using a pastry bag we recommend using a drinking glass so that you have both hands available to fill the bag.
